When in the Dolatshahi lab, Dr. Anne Talkington’s research was focused on studying the mechanisms of interaction between tumor and immune cells, with the goal of understanding how these interactions correspond to treatment resistance in cancer. She used a combination of mathematical modeling and data-based methods to associate tumor-immune interaction networks with the characteristics of a patient's tumor. She aimed to understand interactions in the tumor microenvironment that could be better targeted to improve responsiveness to therapy, and thus, patient outcome. Prior to joining UVA, Anne completed her PhD in Computational Biology at UNC Chapel Hill, where she used a combination of physiologically-based pharmacokinetic modeling and experimental techniques to study adverse responses to PEGylated therapeutics as a result of anti-PEG antibodies. She previously worked on differential equation models for immunotherapies as an undergraduate at Duke University. Tumor-inhibiting and tumor-promoting tumor-immune interactions have been identified as prognostic markers and immunotherapeutic targets such as checkpoint inhibitors. The prognostic and therapeutic success of these solutions, however, have been limited likely due to the highly complex stromal-tumor-immune interactions present in the tumor microenvironment (TME). The cellular interactions in the TME are mediated by ligand-receptor interactions, the complexities of which are further multiplied by the diverse post-translational modifications (PTM) of these secreted and cell-surface proteins. Glycosylation is one of the most common types of PTM and a critical determinant of the function of proteins, namely immune factors, such as immunoglobulins (Ig), tyrosine kinase receptors, integrins, and several other receptors and cytokines. Alterations in glycosylation are common in various cancer contexts and vary by disease progression. While protein glycosylation and TME alterations are known cancer hallmarks, a combined systems analysis of these inter-related changes is lacking. Accordingly, the overall goal of this project is to identify novel tumor-inhibiting or tumor-promoting interactions that are influenced by or impact glycosylation. In a collaborative systems serology project, we set out to define the rules that govern transfer of antibody functionality across placenta from mother to child. We examined paired maternal and cord blood of a cohort of mothers and children for their antigen-specific immune profiles including: (a) antibody subclasses, (b) glycan profiles and the incurred antibody dependent phagocytosis by neutrophils and monocytes, (c) cytotoxicity and secretion of cytokines and chemokines by natural killer (NK) cells, and (d) complement deposition. Using Partial Least Squares Discriminant Analysis (PLSDA), we first defined the top contributors to the immune profile differences between moms and infants, including NK cell functions, IgG1 and doubly-galactosylated IgG. These technologies provide multi-modal snapshots of biological systems at a range of spatial and temporal scales. To weave mechanistic stories based on a multitude of intracellular molecular measurements, inter-cellular, tissue-level and functional data, sophisticated computational methods are required. To this end, we combine multiplex experimental measurements with computational methods to solve problems in the context of cancer, infectious disease and maternal-neonatal immunology among others. He received his Ph.D. in BME in July 2025. His thesis work in the Dolatshahi Lab was titled “Systems Biology Approaches to Improving Immunotherapeutic Outcomes in Solid Tumors“. Rob grew up in Atlanta, GA and Nashville, TN, and graduated from University of Georgia in Athens, GA in 2015 with a major in Biochemical Engineering. Prior to joining UVA, Rob worked for Juno Therapeutics in Seattle, WA as a process engineer. He was a member of the Viral Vector Process Development team and helped design adherent and suspension lentiviral production processes for use in CAR-T therapies. Rob’s research focuses on immune cell behaviors in solid tumors. His work explores how the tumor microenvironment changes T cell and macrophage interactions with targeted immunotherapies. His approach integrates single-cell transcriptomic analysis, mechanistic modeling, and genetic engineering to better understand cell-drug interactions. His projects have included an investigation into the dynamics of T cell inhibitory receptor expression, glycosylation-driven interference in macrophage-antibody interactions, and mechanisms of tertiary lymphoid structure development by cancer-associated fibroblasts. As a postdoc, Rob is working on furthering our understanding of macrophage glycosylation on antibody binding. He hopes to use his findings to better design antibodies to initiate immune reactions in solid tumors. Additionally, he is applying his expertise to help build models of antibody transfer across the placenta. She grew up in Blacksburg, VA, and graduated from Michigan State University in 2022 with a major in Chemical Engineering. She is passionate about teaching and hopes to pursue an academic career path. She is currently co-instructing and designing a new course about Systems Immunology and Engineering with Sepideh. Shay's research focuses on the role of glycosylation in cancer immunology. Specifically, she is studying the regulatory mechanisms surrounding the abnormal patterns of glycosylation that are frequently observed in cancer, as well as how these patterns influence interactions with the immune system and are affected by hypoxia. Understanding the transcriptional, epigenetic, and environmental factors that influence glycosylation could help to improve the efficacy and targeting of immunotherapies. He received his Ph.D. in BME in Summer 2025. His thesis work in the Dolatshahi Lab was titled “Integrative Spatial and Transcriptomic Profiling of Tumor-Immune Interactions in Lung Cancer“. Gabe is from Duluth, Minnesota, and graduated from Saint John’s University (MN) in 2016 with a degree in Applied Physics. Prior to joining UVA, Gabe spent a year living in monasteries in South Asia, serving as a volunteer in the Benedictine Volunteer Corps, and upon his return, worked as a project engineer at Open Systems International, where he designed and implemented multi-server supervisory control and data acquisition systems for power utility companies. Gabe’s research is centered on understanding the coordination and communication of the immune system. His main project focuses on how immune interactions are rewired in lung cancer and how to better leverage the immune system to improve cancer treatment. He is also applying this framework to understand immune regulation and cross-talk at the maternal-neonatal interface. Outside of the lab, you can find Gabe backpacking and hiking through the mountains near Charlottesville, playing the guitar, or tending to one of the many plants in his garden. Student UVA Engineering School Distinguished Fellow (2023-2024) Systems and Biomolecular Data Science Trainee (2024-2025) Mary completed her M.Sc. studies in the Department of Biomedical Engineering in Summer 2025. She grew up in Colonial Beach, Virginia and graduated from Randolph-Macon College in 2023 with a major in Engineering and minors in Chemistry and Math. Mary’s research focused on cancer immunology. Specifically, she was looking at how immune checkpoint inhibitor (ICI) therapies may impact cell-cell crosstalk in the tumor microenvironment, which could help identify patient prognostics and suggest novel targets for supplementary immunotherapies. She was eager to join the team to aid with clinical trials related to tracking antibodies and other markers in maternal-neonatal immunology and to be of assistance in supporting the lab. Abigail grew up across three different states (VA, GA, and SC) but claims Charlottesville as her home. As one of seven siblings, she enjoys spending time with family and loves being an aunt. She joined the Dolatshahi Lab in July 2024 as part of the BioLAB traineeship program. During her traineeship, Anna is contributing to a project focusing on maternal-neonatal immunology. Her primary task is to design a genetically modified trophoblast cell line that overexpresses the antibody receptor FCGR3A. This will allow her to explore the receptor's role in IgG transcytosis during the transplacental antibody transfer in pregnancy. In her free time, Anna enjoys sports, traveling, and spending time with friends. She grew up in Western Massachusetts and graduated from the University of Rhode Island with a degree in Pharmaceutical Sciences with a minor in Biological Sciences. Caitlin previously worked at various Biotech startups in Boston, supporting pre-clinical studies and assay development. Her work focused primarily on cancer vaccines and drug development for rare diseases of mineralization. As a part of the Systems Immunology Lab at UVA, Caitlin's research focuses on maternal-neonatal immunology. Specifically, utilizing systems biology approaches to investigate how social determinants of health and demographic factors affect the placental and systemic immune landscape during pregnancy. Outside of the lab, Caitlin enjoys connecting with long-distance friends through video games and spending time with her local friends (and their pets). At Holy Cross, she was also a member of the Varsity Women's Soccer team. In the Systems Immunology Lab at UVA, her research focuses on fetal-maternal health, with a particular interest in understanding how social determinants of health and clinical factors influence immune response and architecture at the maternal-fetal interface during pregnancy.
